The county's Fire and Rescue said they were called to an "explosion" at a property in Valley View Road, in Riddings near Alfreton, shortly after 07:30 BST. It is thought there could be people inside the property in Valley View Road, in Riddings near Alfreton.
Officers suspect there are people inside the damaged house. The alarm was raised shortly after 07:30 BST after neighbours heard what they told the BBC was a "bang".
Fire crews have been unable to enter the building but East Midlands Ambulance Service have sent their Hazardous Area Response Team (HART).
A police spokeswoman said it was far too soon to say what caused the explosion and subsequent fire.
Gas has not been ruled out as being the cause, she said.
