A spokesman said they had treated three casualties, one of them serious, and the "walking wounded". John Dodsworth said: "It's all gone crazy in town. Someone said a lady was under the bus."
Durham Police is asking people to avoid the area. A force spokesman said: "The bus left the road - we don't know why - and has crashed into a Halifax."
There is a road closure in place from St. Cuthbert's Way roundabout to Crown Street and a pedestrian cordon from Marks and Spencer to the pedestrian zone at the top of Crown Street. The ambulance service said three casualties had been treated - one of them serious - and paramedics were tending to the "walking wounded".
"We have sent the air ambulance, three double-crewed ambulances and a hazardous area rapid response team. Two patients have left for hospital," a spokeswoman added.
County Durham and Darlington Fire and Rescue said it could not confirm if the injured people were pedestrians or bus passengers.
Shops near to the crash, between the Northgate and Crown Street junctions, have been closed and police officers are moving bystanders.
