Firefighters were called to United Molasses on the quayside of King George Dock at 22:15 BST on Monday. There are concerns about a vapour cloud so fire crews are spraying a fine water mist to stop it spreading.
The "significant" leak, at the east side of the dock near the Northern Gateway, led to people in the area being advised to close windows and doors as a precautionary measure. Twenty five firefighters were sent to United Molasses on the quayside of King George Dock at 22:15 BST on Monday.
Humberside Fire and Rescue Service contained it by 04:20 BST. The leak from the tank has been stopped, but an exclusion zone is in place within the port, closing the eastern access road.

Associated British Ports (ABP) said it was assisting the fire service and other authorities "to resolve the situation as quickly as possible".
A spokesperson said: "An exclusion zone within a section of the Port of Hull has been established.
"Please be assured nobody outside the exclusion zone should be concerned."

Simon Donnachie, station manager at Humberside Fire and Rescue Service, said: "With any sort of corrosive leak you're going to have some sort of vapour cloud.
"Due to the time of the day, the location and the direction of wind, there were no real issues to public health."
The Environment Agency said it attended in the early hours of the morning, but there currently appeared to be no leak into the Humber or any other environmental impact.
