The crash forced the closure of the road in both directions but it has since been reopened. He landed on a bank about 30ft (9m) down from the bridge, close to lane one of the eastbound carriageway.
Forensic officers are carrying out investigations on the flyover, which has been reopened, police said. The man suffered "serious head and leg injuries" and was taken to Derriford Hospital.
The man driving the car was also injured and taken to the same hospital. Details of the injuries are not known.
