The road is currently closed in both directions between Swanley and junction two for the M20 interchange. Police said a man, 63, and a woman, 78, who were in the Mercedes C200 car, and a man, 48, who was a passenger in the Toyota Hilux pickup truck, all died.
Police are at the scene but no other details have been released. A man who was driving the Toyota and his two women passengers were taken to Maidstone hospital with serious but non life-threatening injuries.
Appealing for witnesses, officers said the Mercedes was travelling up Wrotham Hill when it was in collision with the Toyota which was travelling in the opposite direction at about 1600 GMT.
The road was closed between Swanley and junction two for the M20 interchange for several hours while police investigated the incident.
It reopened on Monday at about 0700 GMT.
None of the people who died have been identified.
