"It was a suicide blast," the city's criminal investigation department chief, Alishah Paktiawal, told the French news agency AFP.
"The casualties are high, both dead and wounded, but we don't know exact figures because the victims have been rushed to different hospitals," he said.
Most of the victims were thought to be interior ministry employees and police officers, he added.
The explosion took place at about 0800 local time (0330 GMT) on a narrow dirt road which has security gate giving access to the ministry building on it.
Several shops were damaged in the blast, witnesses said.
There was no immediate claim of responsibility for the blast but attacks by Taleban militants have been increasing in recent weeks.
In the capital earlier this month a suicide bomber struck outside the compound housing the US embassy, killing at least 16 people, two of them American soldiers.
