A spokesman for Strathclyde Police said: "Police assisted fire crew to evacuate the majority of residents, five people were still trapped inside and fire services brought them out. Seven residents have been taken to Hairmyres Hospital."
A total of 16 fire crews attended and an investigation into the cause was getting under way.
BBC Scotland reporter Laura Pettigrew said: "There are a number of worried relatives at the police line and I spoke to a man whose grandmother is in the care home and he is extremely concerned.
"He's now spoken to people in the home and the emergency services who have put his mind at rest.
"They said that all of the residents are now out of the home and most of them are safe and well."
A fleet of patient transport minibuses has arrived at the scene to take the pensioners to alternative accommodation.
South Lanarkshire Council said it would provide temporary accommodation for residents, if required.
