Devon and Cornwall Police said next of kin have been informed. The fire is not being treated as suspicious. A six-year-old boy and a 36-year-old man were treated in hospital for the effects of breathing in smoke, but were later discharged. The fire is not thought to be suspicious.
Eight people were in the house - seven members of a family and another person - when the fire broke out just before 08:00 BST.
Devon and Cornwall Police said next of kin had been informed.
A police statement said: "It is with sadness that we can report that a 17-year-old female and an 18-year-old male have now been pronounced deceased as a result of injuries sustained during the house fire at Lee Close, Honiton yesterday morning.
"Our thoughts are with family at this time and the investigation to establish the cause of the fire is ongoing."
