The route N38 bus and the car collided on Lea Bridge Road, at its junction with Essex Wharf, in Clapton, at about 02:20 BST, the Met Police said. The man who died has been named locally as Delano Moore.
The man who died and the man said to have been severely injured, were in the car. He and the man said to be severely injured were in the car with two other men.
Two other men in the back of the car, are in a serious but stable condition. The route N38 bus was in collision with the car on Lea Bridge Road, at its junction with Essex Wharf in Clapton, at about 02:20 BST, police said.

Arriva, which operated the bus involved in the collision, said the driver was treated in hospital after suffering cuts and bruises and was later released.

A London Ambulance spokesman said: "Staff treated 13 patients - three men, reported to be in their 20s, one for an arm injury, the second for a head injury and the third for an arm and head injury. There were road closures in place from Chatsworth Road with the junction of Lea Bridge Road to Orient Way, at the junction with Lea Bridge Road, but all roads have now reopened.
