The two children in the house at the time of blaze were taken to the Victoria Hospital in Kirkcaldy, but later transferred to the Sick Children's Hospital in Edinburgh.
An NHS Fife spokeswoman said they were in a "satisfactory" condition.
The surviving adults are being treated at the Queen Margaret Hospital in Dunfermline.
The spokeswoman said one was in a stable condition and the other was comfortable. The condition of the third adult is not known.
