Police said the block of flats, on which the crane collapsed, was located adjacent to a construction site.
The building has been evacuated by the police. No one has been injured there, they said.
Police have yet to identify the victims and inform their next-of-kin.
Local resident Ron Brown said, "I just heard a massive bang and I thought a bomb had gone off. Then I saw loads of dust go up.
"When I got round to where it had happened I saw the front end of the crane had hit a block of flats and before I knew it the emergency services were crawling all over the place," he said.
The crane was approximately 50 metres high, police said.
A man who suffered minor head injuries has been taken to St Thomas's Hospital.
Eight ambulances, one air ambulance, two motorcycles and a fast response car were sent to the scene.
Nine Elms Lane in Battersea has been closed both ways between Vauxhall and Battersea Park rail station.
