A North West Ambulance spokeswoman said two other people were also taken to Manchester Royal Infirmary with less serious burns to their arms and legs. No details have been released about the extent of the fourth person's injuries.
Albert Square was evacuated after the explosion, which started a fire which destroyed a market stall at the festival.
A Greater Manchester fire service spokeswoman said: "Three 20kg propane cylinders had become involved in the fire and these had to be isolated by fire service personnel to stop the incident from escalating.
"Three people received burns and were treated at the scene before being taken to Manchester Royal Infirmary."
The Health and Safety Executive is invesigating the incident.
